چکیده
Downlo DNA damage response (DDR) activates downstream pathways including cell cycle checkpoints. The D1 gene is overexpressed or amplified in many human cancers and is required for gastrointestinal, , and skin tumors in murine models. A common polymorphism in the human cyclin D1 gene is atively spliced, resulting in cyclin D1a and D1b proteins that differ in their carboxyl terminus. Cyclin erexpression enhances DNA damage–induced apoptosis. The role of cyclin D1 and the alternative form in regulating the DDR is not well understood. Herein cyclin D1a overexpression enhanced the s characterized by induction of γH2AX phosphorylation, the assembly of DNA repair foci, specific tment of DNA repair factors to chromatin, and G2-M arrest. Cyclin D1 deletion in fibroblasts or interfering RNA–mediated reduction of endogenous cyclin D1 in colon cancer cells reduced the small 5-fluorouracil–mediated DDR. Mechanistic studies showed that cyclin D1a, like DNA repair factors, elicited the DDR when stably associated with chromatin. Cancer Res; 70(21); OF1–10. ©2010 AACR.

Downlo roRNAs (miRNA), a class of natural RNA-interfering agents, have recently been identified as attractive s for therapeutic intervention. The rationale for developing miRNA therapeutics is based on the premise berrantly expressed miRNAs play key roles in the development of human disease, and that correcting miRNA deficiencies by either antagonizing or restoring miRNA function may provide a ...

Downlo atocellular carcinoma (HCC) is generally a fatal disease due to a paucity of effective treatment options. entification of oncogenic microRNAs that exert pleiotropic effects in HCC cells may offer new theratargets. In this study, we have identified the human microRNA miR-191 as a potential target for HCC y. Inhibition of miR-191 decreased cell proliferation and induced apoptosis in vitro ...

متن کاملThe AC133 epitope, but not the CD133 protein, is lost upon cancer stem cell differentiation.
Colon cancer stem cells (CSC) can be identified with AC133, an antibody that detects an epitope on CD133. However, recent evidence suggests that expression of CD133 is not restricted to CSCs, but is also expressed on differentiated tumor cells. Intriguingly, we observed that detection of the AC133 epitope on the cell surface decreased upon differentiation of CSC in a manner that correlated with...
